How pro do you want it?.. My experience is that less is better
I have used Adobe Premiere and Adobe After Effects a lot and every time I wish I would have used Microsoft Movie Maker

I like both Premiere and After Effects.. But for most of my needs.. I'll keep it simple using Movie Maker and iMovie :P
And.. I think most of the awsomeness of a video is within the shot itself
